The e-vouchers will be available at more than 600 of the grocer's shops.
Skype hardware, including headsets and handsets to plug into computers, will also soon be available to buy in stores.
Sainsbury's home services commercial manager Adam Zeiderman said: 'It will complement our existing telecommunications offer. Combined with our experience of selling mobile top-up cards, we anticipate that the system will integrate seamlessly into all our stores.'
Skype UK market development manager Alison Shrimpton said: 'Buying Skype credit with cash while doing your regular shopping makes the whole Skype experience even more accessible to existing users and new ones.'
Ebay-owned Skype allows users to make calls to anywhere in the world over the internet. All voice and video calls to other Skype users are free.
